Just squeeze a bit of silicone cement into the hole and let it cure. Just make sure you seal the correct hole (there are two). As for function, the hole that's open serves to accentuate the Venturi action in the housing (that's the one facing the mouthpiece). The other one, if left open, will create turbulence and slightly decrease the net total flow directed toward the mouthpiece. It's not critical, but it is noticeable when you adjust the vane. There are two holes for regs that used to use the same valve body, but were reversible for hose placement on the other side.

When you squeeze in that dollop of silicone goop, make sure to leave a little extra on the outside (like a round-head screw head). That'll keep it from getting pushed out of the valve body and blown out the mouthpiece when you purge the reg one day. That extra goop nestles up against the case, to keep the plug in place.
