Assuming I elevate my heart rate to target zone while cardio-ing is it preferable to elevate via "intensity" (say walking at 4+ MPH on an inclined treadmill) or speed (run on level treadmill)
 
Your heart and lungs don't make a distinction like that. It's your heart rate and respiratory rate that matters, and for most people running gets the heart rate up higher. However, if you have your heart rate in a lower threshold when walking, that just means you have to exercise for longer to burn the same amount of calories.

But it depends on your goals. Are you training for endurance competitions or just trying to lose weight?

Honestly, cross-training so you don't plateau is the best. Interval training (intersperse of sprints and active recovery) is one of the best ways to increase your anaerobic threshold.

Also make sure you are doing resistance training to build muscle mass - the higher your lean body ratio is the higher your metabolism will be.
 
We don't really understand exercise physiology all that well, so no one can answer your question.

It seems intuitively obvious that there is a difference, and there is probably some relative difference in the way your body reacts over different types of muscle fibers. But which is "better for you" is not a question we can answer today.

That said, if you're just trying to lose weight, interval training is the big "thing" for that result these days. While no one has done any significant, controlled, longitudinal studies on the outcomes of various interval methods versus just staying in a target range, the short term results appear promising.
