If I attach the hoses to the first stage for the very first time ( after I remove the plug ) should I but some silicone grease on the O-ring or are they OK to go out of the box?
 
But I should lube them up?
 
This is one of those never ending debates. My take in it is this. Since the reg and orings are new there is no chance of residual grease on them from a prior assembly so I put a SLIGHT amount of grease on them. While the orings are static is service (don't more) and don't need to be lubed, they are technically dynamic while they are being installed ( and those who are starting to type they are not, please go read the Parker Oring manual first) it's not a bad idea to have a little lube on them, at least on the initial installation. For subsequent assemblies the residual lube is enough. What to use is another matter of opinion. Silicone is fine, so is Christolube and frankly spit will work it that is all you have.

because a thin film of lubricant will prevent the threads from being stuck (corrosion). In particular the ports that are unused. I had one that gave me a REAL HARD TIME because it was stuck solid.
 
I put a little bit of lube on the o-rings just to help with getting them over the threads. If I don't have christolube handy, I don't worry too much about. Also when tightening down the hose, it needs to be nug, not hercules tight.
 
Scuba is an activity where spitting is socially acceptable and encouraged at times :)

Seriously, I usually like to put just a tiny bit of Tribolube on hose o-rings before installing (or I use spit in a pinch).
