We are looking to rent a room and a boat for a long weekend on Oct 12th-15th near Big Pine Key. Any suggestions? Looking for typical center console 21-23ft boat. It's 2 couples, so a 2 room suite or something like that would be great.

http://www.parmersresort.com/ is a possibility. Has boat ramp and dock onsite. Little Torch Key, just south of Big Pine Key, around MM28. Only if Parmer's is booked would I consider the Big Pine Key Fishing Lodge, which is at the very north end of Big Pine Key. Really a trailer park with a few rooms, but it does have docks right next to the motel section.

I don't have info on boat rentals, but another alternative is Wreckreation Charters, a 6 passenger dive boat running out of Cudjoe Gardens Marina, MM21.

Or the owner/operator of Wreckreation Charters, on SB as "Biscuit7" could probably point you towards some local boat rental companies.
 
Try Dolphin Marina for boat rentals.Boats are relatively nice.They have a website somewhere.
We stayed in Little Torch for quite a few years and loved it.

The Busch and Looe Key are great dives and the bugging is better than the Upper Keys.Inshore of Hawk Channel vis runs 10' to 30' and offshore to the reef runs 30' or better normally.
 
The least expensive hotel in the keys is the Holiday Inn on Marathon at about $68/night
