In NITROX mode, aftering entering the % O2 value, how long does one have to commence the dive before the computer defaults to 21% ?

Published time is 2 hours if you have no dives on it that day (that is, first dive in a series)
It will not default back to 21% until the expiration of desat time if you have started a dive series.
I set it while gearing up for the first dive of the day, and check it before jumping in on subsequent dives (particularly since I frequently dive a cut mix of 30% or so for deeper dives, and 32% for shallower ones, and sometimes will finish a day with a tank of air if we're going to be shallow and I'm being cheap
)
