How is the camera connected to the strobe? Via a cord or wire, is there are anything between the camera and the strobe? Controllers maybe? Can you connect the camera directly to the strobe via just one cord?

The DS-50 in Manual or TTL does not matter, it will still fire in sync with the camera as long as the cord is making the connection.

The strobe is connected with a 4 prong cable and internal to the camera housing is connected to the hot shoe of the camera. I thought that it was working OK out of the caverns, but perhaps I was mistaken.

Check this at Ikelite on testing your strobe:

Ikelites TTL cable should be 5 pin, I believe. Are you sure its 4 pins?
