Can a manifold be taken off a set of twin tanks when they are full? I have a set of doubles filled with Nitrox that I would like to convert back to single tanks. The manifold has a plug that can screw into the end when the manifold is removed but I am not sure if I will need to drain the tanks before disassembly of the crossbar/isolation valve.

NO, DO NOT take the manifold apart with pressure in the tanks. The crossbar/isolator connects the tanks before the valves and is always pressurized.

Can you? Maybe but you and anyone around you may not like the results:) Like Lobzilla said, the manifold is always under pressure. Your handwheel controls air to the outlet(regulator) but not to the manifold.
