I have a balanced diapragm regulator that i want to take apart, but the diaphragm clamping part does not loosen from the main body using a spanner.
Have you any suggestions?

Make the expansion and contraction of metals exposed to hot and cold work for you. Resorting to excess force will be conclusive but you may not like the result. Hot and cold will do the job eventually unless the job is hopeless.
 
Could try putting it in a ultrasonic cleaner for a while and see if that breaks some of the crud up.

More force usually works but finesse is safer.
