My guess would be Brisbane-Cairns-Guam-Palau.

I think that is right. My wife will be traveling from Perth to Palau on that route. She did a fair amount of research last week re cost/travel time. We are meeting in Guam for the trip to Palau. I can't wait.
 
We are staying at PPR and diving with Neco Marine. NM was recommended be a friend and highly rated on the internet. Liveaboards are supposed to be better for diving, but we generally prefer to be land based. Our next liveaboard will probably be at Wakatobi.
 
Another option may be brisbane to Manila to palau. I think Philippine airlines now fly from Brisbane to Manila via Darwin.
I was on the big blue explorer for 5 days followed by 3 days land based with sams. If I was going back I would try to get on the siren boat.
 
As others have said, last time I looked at this trip from Australia, you had to go through Guam or Manila. Problem for me was if I got to Manila I would be too tempted to go diving in the Philippines. Also the flights schedules were pretty bad in terms of stopovers/late arrival/early delartures, basically lost another 2 days out of the trip to go on to Palau
